Dolly Parton talks dreams, love, plastic surgery

NEW YORK (Reuters) - Although Dolly Parton has cemented her place in country and popular music, pop culture, and as an entrepreneur and philanthropist, she still, on occasion, gets nervous.

Her new book, "Dream More: Celebrate the Dreamer in You" encourages readers to overcome their fears, believe in their passions and keep taking risks.

The "I Will Always Love You" singer/songwriter, 66, who has written more than 3,000 songs and sold more than 100 million records, talked to Reuters about the message of the book, which was published this week.

Q. You say you put off writing this book?

A. "It's just a simple little book. It's not meant to save the world, or it's not a complete book of how to be successful, but I think there is enough stuff in it for people to see kinda how I conduct my business and kinda what my thoughts are. And the good part is that all the money, if it sells good, goes to Imagination Library."

Q. Right - your nonprofit quest to get kids to read?

A. "It's one of the reasons I wanted to write this too, because I usually do concerts every year, for the foundation to make money to afford a lot of books, but I am not on tour now."

Q. Talk about your 2009 commencement address at the University of Tennessee. Were you nervous?

A. "Well, yes, when I am out of my element doing things. I am not that educated and I didn't go that far in school and I thought, 'What am I going to say to these educated people, not just these kids who have just graduated college and are probably brilliant, but all these professionals and all these teachers?' And I thought, 'Oh, I am not smart enough', but I thought, 'Well, at least I am a hometown girl. At least they can see that in America, you can start from humble beginnings, that everybody can make it."

Q. Which is one of the book's messages, overcoming fears?

A. "Any time I am in a situation where I am just not comfortable, I am uneasy, but that doesn't mean I won't go on with it, just like the speech. And that I won't be good at it, but there are just some things I would prefer not to do!"

Q. Success doesn't equal happiness, yet you seem so hopeful and modest?

A. "I am always hopeful as a person, I have been since I was little...I really want things to be good. As I mention in the book, I wake up everyday expecting it to be good, and if it is not, then I try to set about changing it before I go to sleep at night."

Q. Would you describe yourself as religious or spiritual?

A. "Just spiritual, I am not religious. Although I grew up in a very religious family, but...I am no fanatic by any stretch of the word, and I am no angel, believe me. I wrote a song called 'The Seeker' many, many years ago, and it says 'I am a seeker, just a poor sinful creature, there is no one weaker than I am.'

"People say, 'What do you regret?' I say, 'I can't say that I regret anything because at the time I was doing it, whatever it was, it seemed to be the thing to be doing at the time.'

"I have a good friend base, I have a good husband. So I have a lot of things and people who help me and guide me. I have never had to go to a psychiatrist, but I would if I thought that I needed to.

Q. But we are in New York, Dolly! No psychiatrist?

A. "Well yes (laughs), I guess not. But I do that in my songs, I write my feelings out and then I have such a strong faith and then I have such good friends. I am very close to several of my sisters, and we just talk about everything and anything....And my best friend Judy, there is nothing I can't tell her, even if it is the awful-est thing in the world."

Q. You recently had to deny gay rumors. Who is your greatest love?

"My husband is my greatest love, I have been with him 48 years...He is my best buddy."

Q. Why do you think people always wonder about him?

A. "They don't think he really exists! When I was doing my show, we were thinking about having a different guy knock on the door every night, as my husband, and then one night he would be a midget, and one night he would be a black man, and one night he would be like a boxer or a wrestler, all these different things that people imagine what my husband looks like."

Q. You say that looking so artificial works for you, as it lets you prove how real you are. Why all the plastic surgery?

A. "Because I need it. Why does anybody get it?"

Q. Why do you think you need it?

A. "Because I am in show business. I am not a natural beauty. And I am on camera all the time. And I just always see, like if I need - Oh take one of my chins off, at least! - Or whatever. I mean, I don't go to extremes with it. I just do little bits and pieces, just to try and keep things touched up, just tweaking."

Precisely engineering 3-D brain tissues

ScienceDaily (Nov. 29, 2012) ? Borrowing from microfabrication techniques used in the semiconductor industry, MIT and Harvard Medical School (HMS) engineers have developed a simple and inexpensive way to create three-dimensional brain tissues in a lab dish.

The new technique yields tissue constructs that closely mimic the cellular composition of those in the living brain, allowing scientists to study how neurons form connections and to predict how cells from individual patients might respond to different drugs. The work also paves the way for developing bioengineered implants to replace damaged tissue for organ systems, according to the researchers.

"We think that by bringing this kind of control and manipulation into neurobiology, we can investigate many different directions," says Utkan Demirci, an assistant professor in the Harvard-MIT Division of Health Sciences and Technology (HST).

Demirci and Ed Boyden, associate professor of biological engineering and brain and cognitive sciences at MIT's Media Lab and McGovern Institute, are senior authors of a paper describing the new technique, which appears in the Nov. 27 online edition of the journal Advanced Materials. The paper's lead author is Umut Gurkan, a postdoc at HST, Harvard Medical School and Brigham and Women's Hospital.

'Unique challenges'

Although researchers have had some success growing artificial tissues such as liver or kidney, "the brain presents some unique challenges," Boyden says. "One of the challenges is the incredible spatial heterogeneity. There are so many kinds of cells, and they have such intricate wiring."

Brain tissue includes many types of neurons, including inhibitory and excitatory neurons, as well as supportive cells such as glial cells. All of these cells occur at specific ratios and in specific locations.

To mimic this architectural complexity in their engineered tissues, the researchers embedded a mixture of brain cells taken from the primary cortex of rats into sheets of hydrogel. They also included components of the extracellular matrix, which provides structural support and helps regulate cell behavior.

Those sheets were then stacked in layers, which can be sealed together using light to crosslink hydrogels. By covering layers of gels with plastic photomasks of varying shapes, the researchers could control how much of the gel was exposed to light, thus controlling the 3-D shape of the multilayer tissue construct.

This type of photolithography is also used to build integrated circuits onto semiconductors -- a process that requires a photomask aligner machine, which costs tens of thousands of dollars. However, the team developed a much less expensive way to assemble tissues using masks made from sheets of plastic, similar to overhead transparencies, held in place with alignment pins.

The tissue cubes can be made with a precision of 10 microns, comparable to the size of a single cell body. At the other end of the spectrum, the researchers are aiming to create a cubic millimeter of brain tissue with 100,000 cells and 900 million connections.

Answering fundamental questions

Because the tissues include a diverse repertoire of brain cells, occurring in the same ratios as they do in natural brain tissue, they could be used to study how neurons form the connections that allow them to communicate with each other.

"In the short term, there's a lot of fundamental questions you can answer about how cells interact with each other and respond to environmental cues," Boyden says.

As a first step, the researchers used these tissue constructs to study how a neuron's environment might constrain its growth. To do this, they placed single neurons in gel cubes of different sizes, then measured the cells' neurites, long extensions that neurons use to communicate with other cells. It turns out that under these conditions, neurons get "claustrophobic," Demirci says. "In small gels, they don't necessarily send out as long neurites as they would in a five-times-larger gel."

In the long term, the researchers hope to gain a better understanding of how to design tissue implants that could be used to replace damaged tissue in patients. Much research has been done in this area, but it has been difficult to figure out whether the new tissues are correctly wiring up with existing tissue and exchanging the right kinds of information.

Another long-term goal is using the tissues for personalized medicine. One day, doctors may be able to take cells from a patient with a neurological disorder and transform them into induced pluripotent stem cells, then induce these constructs to grow into neurons in a lab dish. By exposing these tissues to many possible drugs, "you might be able to figure out if a drug would benefit that person without having to spend years giving them lots of different drugs," Boyden says.

Stocks edge higher as budget talks move ahead

(AP) ? Optimism that a budget deal will be reached in Washington helped lift the stock market in early Thursday trading. A pair of economic reports also brightened the mood.

A half-hour after the opening bell, the Dow Jones industrial average was up 50 points at 13,034. UnitedHealth Group led the 30 stocks in the Dow, rising 66 cents to $53.43.

Lawmakers are negotiating a deal to avoid the "fiscal cliff," a collection of tax hikes and federal spending cuts set to start Jan. 1. Later Thursday, Treasury Secretary Timothy Geithner will meet with Senate Republican Leader Mitch McConnell and Senate Democratic Leader Harry Reid to push the negotiations forward.

In other trading, the Standard & Poor's 500 index rose seven points to 1,416 while the Nasdaq composite gained 19 points to 3,011.

The Commerce Department raised its estimate for U.S. economic growth to an annual rate of 2.7 percent in the July-through-September period. That's much better than the 2 percent rate estimated a month ago and more than twice the 1.3 percent rate logged in the three previous months.

Growth during the end of the year is likely to take a hit from Superstorm Sandy, along with worries that Congress won't avoid falling over the fiscal cliff. Economists estimate that growth is slowing to a rate below 2 percent in the current October-December period.

The Labor Department also reported that the number of Americans applying for unemployment benefits dropped to 393,000 last week, in line with what economists had expected. It was the second straight drop after Superstorm Sandy drove applications higher earlier this month.

Some retail stores posted poor sales numbers, driving their stocks lower. It's a critical time for retailers, who log a huge chunk of their yearly profits in the weeks running up to the holidays.

Kohl's plunged $5.41, or 10 percent, to $45.74, the most in the S&P 500 index. The company posted a drop in sales and said stores in the Mid-Atlantic and the Northeast, areas hit by Superstorm Sandy, fared the worst. Results at Target, The Gap, and others also fell short of Wall Street expectations.

Kroger Co. rose 82 cents to $25.88 after the supermarket chain reported stronger quarterly profits and raised its earnings outlook for the year. Stronger sales helped the operator of Fred Meyer and Food 4 Less stores post better results than analysts had expected.

Myanmar cracks down on mine protest; dozens hurt

(AP) ? Security forces used water cannons and other riot gear Thursday to clear protesters from a copper mine in in northwestern Myanmar, wounding villagers and Buddhist monks just hours before opposition leader Aung San Suu Kyi was to visit the area to hear their grievances.

The crackdown at the Letpadaung mine near the town of Monywa risks becoming a public relations and political fiasco for the reformist government of President Thein Sein, which has been touting its transition to democracy after almost five decades of repressive military rule.

The environmental and social damage allegedly produced by the mine has become a popular cause in activist circles, but was not yet a matter of broad public concern. But hurting monks ? as admired for their social activism as they are revered for their spiritual beliefs ? is sure to antagonize many ordinary people, especially as Suu Kyi's visit highlights the events.

"This is unacceptable," said Ottama Thara, a 25-year-old monk who was at the protest. "This kind of violence should not happen under a government that says it is committed to democratic reforms."

According to a nurse at a Monywa hospital, 27 monks and one other person were admitted with burns caused by some sort of projectile that released sparks or embers. Two of the monks with serious injuries were sent for treatment in Mandalay, Myanmar's second biggest city, a 2 ? hour drive away. Other evicted protesters gathered at a Buddhist temple about 5 kilometers (3 miles) from the mine's gates.

Lending further sympathy to the protesters' cause is whom they are fighting against. The mining operation is a joint venture between a Chinese company and a holding company controlled by Myanmar's military. Most people remain suspicious of the military, while China is widely seen as having propped up army rule for years, in addition to being an aggressive investor exploiting the country's many natural resources.

Government officials had publicly stated that the protest risked scaring off foreign investment that is key to building the economy after decades of neglect.

State television had broadcast an announcement Tuesday night that ordered protesters to cease their occupation of the mine by midnight or face legal action. It said operations at the mine had been halted since Nov. 18, after protesters occupied the area.

Some villagers among a claimed 1,000 protesters left the six encampments they had at the mine after the order was issued. But others stayed through Wednesday, including about 100 monks.

Police moved in to disperse them early Thursday.

"Around 2:30 a.m. police announced they would give us five minutes to leave," said protester Aung Myint Htway, a peanut farmer whose face and body were covered with black patches of burned skin. He said police fired water cannons first and then shot what he and others called flare guns.

"They fired black balls that exploded into fire sparks. They shot about six times. People ran away and they followed us," he said, still writhing hours later from pain. "It's very hot."

Photos of the wounded monks showed they had sustained serious burns on parts of their bodies. It was unclear what sort of weapon caused them.

The protest is the latest major example of increased activism by citizens since the elected government took over last year. Political and economic liberalization under Thein Sein has won praise from Western governments, which have eased sanctions imposed on the previous military government because of its poor record on human and civil rights. However, the military still retains major influence over the government, and some critics fear that democratic gains could easily be rolled back.

In Myanmar's main city of Yangon, six anti-mine activists who staged a small protest were detained Monday and Tuesday, said one of their colleagues, who asked not to be identified because he did not want to attract attention from the authorities.

As it turns out, many other non-Africans have delighted in chimps before me. Since 1950, the small West African country exported more than 2,000 live chimps to the US, Japan, and Europe for the entertainment industry and for biomedical research. A law restricting such exports was passed in the eighties, but chimps continued to be owned within Sierra Leone, typically by tourists and expatriates including Peace Corps volunteers [reference 1]. A young chimp sells for US$61 on average, which is a pretty price in a country where most people survive on less than $1 per day.

Young chimps are super cute! But as they age, the jungle within appears. They flip tables, tear down drapes, and occasionally attack. Thus, in 1995, an accountant in Sierra Leone, Bala Amarasekaran, established Tacugama Chimp Sanctuary to rehabilitate orphaned chimps, and to advocate against the chimpanzee pet trade. Some of the sanctuary?s chimps are donated by expatriates who no longer want their pets. Others are discovered within cages in abandoned houses, or chained by the road for sale. More recently, authorities confiscate illegally owned chimps because of a 2007 law prohibiting the capture, killing, or possession of chimps in Sierra Leone, and bring them to Tacugama.

A skinny Sierra Leonean man with a lazy eye and a warm demeanor named Moses Kappia led my tour of Tacugama Sanctuary. As we watched chimps howl and bounce through various rehabilitation enclosures, we talked about the history of the sanctuary.

When Kappia arrived at Tacugama 16 years ago, he watched it blossom with start-up donations from the European Union. Soon after, the sanctuary struggled to survive as civil war ripped through Sierra Leone. In 1997, rebels broke into the sanctuary and threatened to kill the chimpanzees for food, but Kappia and the few remaining staff convinced them to spare the animals. Instead, the rebels stole their belongings, their food, and the food and medicine intended for the chimps. The rebels returned again during that brutal, 11-year ?blood diamond? war, and again Kappia defended the chimps.

As the war wound down in 2002, Tacugama joined together with other African primate sanctuaries in the Pan African Sanctuary Alliance. They still struggle for funds, but manage to care for nearly 100 orphaned chimps that now frolic in their fenced-in slice of jungle.

Ali, my taxi driver, accompanied me on the tour. Inspired by Kappia, he vowed to report any chimps he discovered being sold or kept. And on the ride home, Ali said that he?d let any ?white people? in his taxi know that he could take them to see (not buy) the chimps ? just a 20 minute drive from the capital city of Freetown.

Back in the US, I?ve taken note of the Captive Primate Safety Act that has languished in Congress since 2007. It seeks to protect monkeys, chimps and other primates from the exotic pet trade in the US. The Humane Society suggests that people urge their legislators to support the act when it is reintroduced next year.

Kappia managed to convince rebels notorious for rape, amputations, and other crimes against humanity to leave the chimps alone. To cross oceans, Hughes hitched lifts with cargo ships. He spent four days in an open fishing canoe from Senegal to Cape Verde, and was then arrested when he arrived.

Later, officials in the Democratic Republic of Congo jailed him for six days believing he was a spy.

?None of this put me off, it just made me more bloody-minded to succeed,? he says.

DEATH IN THE FAMILY

The hardest point, ?when I just wanted to give up,? he remembers, was after his older sister, Nicola, died from cancer two years ago at the age of 39. Hughes rushed home to see her before she died.

?She told me not to stop the trip, but I was at a real low point. I?d done 184 countries and had only 17 to go and I thought why not leave it there,? he says. The memory of his sister spurred him on, as did the people that he met as he traveled and the money he was raising for Water Aid, which works to bring clean water to people in the developing world.

?If you take everything that you know of the world from the news, it?s all the bad stuff and you get very paranoid that everyone is out to get you,? he says. ?But the most amazing thing to me is that everyone I met looked after me and I didn?t even know them.?

Hughes plans to stay in South Sudan only until Wednesday. But he will not then be flying home.

He says to ?keep in the spirit of the adventure? he will continue through Africa and across Europe by bus and boat, aiming to return home to Liverpool by ferry from Ireland in time for Christmas.

?Someone wrote to me and pointed out that this would be the trip of a lifetime for most people, but for me it?s essentially just the bus home,? he says. After a long rest, he says he will then begin exploring options to continue with a career in film-making.

BOJ rift surfaces over easing as political debate heats up

TOKYO/NAGOYA, Japan (Reuters) - A split within the Bank of Japan was laid bare on Monday, with newcomers to its board pressing to strengthen commitment to an ultra-loose policy, as pressure on the central bank for bolder action intensified in the run up to next month's election.

Shifting the debate away from the central bank, BOJ Governor Masaaki Shirakawa repeated his view that monetary easing alone cannot beat deflation, urging the government pursue fiscal reform and deregulation to boost domestic investment.

But minutes of the October 30 meeting of the nine-member board released on Monday showed Takehiro Sato, one the newcomers, had suggested a more activist stance on monetary expansion once Shirakawa's term ends in April.

The conduct of policy became one of the most heated areas of political debate ahead of the December 16 election after Shinzo Abe, the leader of the opposition Liberal Democratic Party and likely next prime minister, called for the central bank to adopt negative interest rates among other radical measures.

Abe also threatened to take away the BOJ's independence to allow the government to call the shots on targets and policy action.

Sato, a former economist who has consistently called for more BOJ measures to keep yen rises in check, proposed linking monetary policy more closely to a rise in consumer prices and suggested giving up the BOJ's view that consumer inflation will approach 1 percent in the fiscal year ending in March 2015.

The minutes of the Oct 30 meeting showed Takahide Kiuchi, another former economist who joined the board with Sato in July, was alone in his support for the proposal, and their views lacked broad support.

"A few members raised the issue of whether it was possible to further exert influence on interest rates and demonstrate the BOJ's clearer stance on monetary easing by changing the current wording," the minutes showed.

"Most members expressed a cautious view regarding making changes to the wording of the policy commitment at this point."

At last month's meeting, the BOJ eased monetary policy by boosting its asset-buying program by 11 trillion yen ($133.5 billion), to 91 trillion yen. It also unveiled a plan to supply banks with unlimited cheap long-term funds under a new scheme initially seen sized around 15 trillion yen.

But that has not eased pressure on the BOJ.

Investors have bet on bolder monetary easing by the BOJ as Abe repeated calls for "unlimited" policy loosening, which drew strong opposition by incumbent prime minister and ruling party leader Yoshihiko Noda.

A weekly gauge of sentiment in the Japanese government bond market worsened to a level unseen in nearly five months on speculation of more aggressive easing following a likely change in the government after an election next month.

The LDP's Abe, who was last prime minister in 2007, is calling for far more aggressive monetary measures to revive Japan's struggling export dependent economy, including a commitment to an inflation target of 2 percent to overcome deflation, with a change to the BOJ's governing law.

Governor Shirakawa declined to respond directly to Abe's requests, saying only that the government and the central bank were doing their best to achieve the 1 percent price goal.

"If efforts to narrow Japan's output gap bears fruit, price growth and price expectations will gradually rise," he told business leaders in Nagoya, central Japan.

Shirakawa also said the BOJ was mindful of the pain a strong yen was inflicting on Japan's economy, suggesting its readiness to ease policy again should Japan slip deeper into recession.

Japan's main opposition Liberal Democratic Party (LDP) kept its clear lead ahead of the election, opinion polls showed on Monday, with the hawkish Japan Restoration Party (JRP) led by a nationalist ex-Tokyo governor running second.
